>>11989031There are 2 ways to make a covid (or any other viral RNA) detection:
1- Direct RTQ PCR without cleaning endogenous cell RNA and DNA: a lot of interferences.
2- The 2-step method which involves eliminating endogenous cell RNA and DNA: less interferences.
Cell mRNA gets amplified when using oligo-T for the retrotranscriptase, so at the end you get both viral and cell cDNA and it makes interferences when using primers for the latter PCR.
You have 2 options:
A- Agar electrophoresis of all amplified material and split the result by size.
B- Use better primers when using the retrotranscriptase, so you won’t make cDNA from cell RNA.
Automatizing this is a bitch.
