It depends on the anime, but overall, I can name a lot more anime and manga which are transgender friendly than I can US cartoons

Its also very GNC and LGBT friendly overall. US cartoons are kinda catching up on the latter but still nowhere close on the former.

You don’t see very many male male relationships outside of niche interests though, but half of moe and slice of life stuff has bisexual/lesbian undertones, from what I’ve seen of those kind of shows a lot don’t even have speaking male characters.
