the joke is a reference to skibidi toilet, in this series toiletmen (men who live in or are part of mobile toilets) are at war with cameramen (men who have cameras and other technology for their heads)
the toilets all sing the song OP first linked and associated with tranime, making it the theme of villains. the speakermen (allies of cameramen) play tears for fears "everybody wants to rule the world" and here OP has associated pepe the frog with the song, and thus, the heroes of skibidi toilet.
what op neglects to realize is that skibidi toilet IS anime, it's a mecha show in disguise
if OP is the type to associate all anime with transexuals (when it's usually just the slice of life and magical girl genres) then he is truly lost
