For one, the "date" was quite obviously not a romantic one if you even watched the closing credits, it was more of a "girls night out" kind of thing that the creators put in the show as a lighthearted joke to close the show off on a positive note. You also can't really take anything Mako says seriously, considering that most things she said in the instance you are referring to are during a "hallelujah" segment.
On the topic of her never showing any interest in men, she does on multiple instances, one of which being the dream sequence she has while wearing Junketsu. She weds a man if you look closely enough. The second example being any time her teacher strips down, she is quite obviously flustered/embarrassed
