When provided proper infrastructure, cars are as convenient as is possible - you can go directly anywhere at any time, right from your front door, by the fastest route available with no excess stops. Transit obviously cannot do this, because it needs to serve large groups as opposed to an individual, it's impossible for even the best transit to be as convenient as a well-supported car. Thus the only way for transit to be the more convenient option is to intentionally handicap car travel, which is clearly stupid.
As for comfort, cars win this one for short-medium distance travel regardless of any circumstance. You have to do the driving yourself, yes, but you're also in a personal space that caters to your every whim with music, air conditioning, reclining seats, and no litter/grime/crackheads, instead of the utilitarian sardine can of a bus or tram. You can make transit more luxurious, but no amount of excessive spending can make it as nice as just having a private, personal space to relax in with no strangers around. Self-driving, should it come to full fruition, absolutely demolishes the one singular argument for the "comfort" of transit.
