For water, terrain, temperature and wind make a huge difference. On some long climbs or with stiff headwinds I've gone through 12+ liters over 100 miles. With a good surface, agreeable temperatures and irrelevant wind, I've done the same distance, drinking only a quarter of that.
Between Richmond and DC you won't need a filter. There are loads of towns. Just stop at a public park or a 7-11 and fill up as often as necessary.
As for the biking itself, I highly recommend shorts with chamois. If you're paranoid about looking like a faggot, wear something over them. It's probably obvious, but pace yourself. As long as you put in the hours, you will cover the miles eventually. You're probably not going to run into serious physical obstacles.
