Generic advice, do not focus on all 100 acres right off the bat. Develop about 5 acres heavily, then the next 5, then the next.
Honestly though it all depends on what you have.
Plains? Raise livestock and plant orchards. Gardening may require more compost depending on the top soil. Hunting may be limited to birds. Watch out for the wind.
Mountains? Try prospecting, there are often minerals you can turn into art. Raise smaller, hardier livestock like alpaca, pigs, or goats. Gardening may require intensive techniques. Hunting is good but takes more energy. Account for the lower oxygen in every breath.
Forest? You now have a wood industry, hope it isn't pine. Learn to carve, craft, and general carpentry. Any crops will likely require clearing some space. Fruit bushes will do well. Hunting is likely excellent. Check the danger of forest fires in that region.
River? Congratulations, you are in a flood zone. Fishing is excellent, hunting is excellent. Livestock is very strong. Crops depend on the soil, better starting chance of good dirt. Flooding is a serious concern, check in the spring to see how upriver melt effects your land.
