Only grown edible mushrooms and haven't messed around with genetics too much but studying mycology is my passion. Will not aid you in your illicit adventures, however can provide you info for purely educational purposes as freedom of information. I will not be responsible for you being retarded, blame your parents for that and unlike your theoretical mushrooms, do not reproduce.
If you were isolating traits of a fungi the easiest would be selective breeding + cloning. Grow the mycellium to fruit, select a fruit expressing traits you like to clone. Take a sample from the inside of the stipe and place on agar until it turns to mycellium, fruit mycellium. Select another fruit you like, this time take a spore sample. Grow those spores into fruit, select a fruit you like, take a sample and clone on agar. Repeat this process a total ~7 times and eventually you will have your own strain.
