Duplo blocks can stack on top of System studs and the standard Duplo block fits a 4x4 space. Some pieces don't quite fit a 4x4 space, so you need to get creative with your build, but if you're just trying to attach some Duplo and System together, the most space-efficient way is to put a 4x4 System plate on the underside.
