It would be similar to the knight/castle themes of the past however Nordic fairytale creatures would be replaced by Greco-Roman mythological beasts.
Regarding the factions, Lego could very easily have them consist of Mediterranids (Greeks, Romans, Carthaginians) vs. monsters vs. Northern Barbarians (Gauls, Germanics, Britons, etc.) They wouldn't have to specifically be Greek or Germanic but you could have general designs inspired by the technology and clothing of the areas historically.
It would also give Lego an excuse to make sets of the Parthenon, Coliseum, or other Classical buildings which they have hitherto shied away from creating. I do know that it is possible to create these sets using minifigures from the Lego Minifigure series, but this is awfully expensive to do.
