Additionally, The Tithes (a GW-produced animated series that was in production WELL before the Twitter post was made) features a female custodian as a main character. So there's 2 supporting pieces of official media.

The fact of the matter is, only Space Marines are stated to exclusively be men. The lore you're thinking of is regarding Space Marine geneseed, which is irrelevant to the issue of Custodes. They are created via "arcane genetic alchemy", and have never been stated to make use of geneseed at all.
The only piece of "lore" regarding their gender is the passage from their first codex stating that "it is a great honor for those of noble Terran houses to submit a son" (to be a candidate for being made a custodian). This wording was changed slightly in 10th edition to say "child" instead of "son", an intentional change which was confirmed by the custodian in The Tithes and their social media team.
But guess what? The first custodes codex came out less than a decade ago, during 7th edition! So no, there are not "decades" of "pre-ceding" lore.
