They would have to make being mute actually be part of his character and have a reason for it, like a wound on his throat, and that's never really been something that characters in the games have drawn attention to.
Since Link to the Past the Zelda games have always had moments where a character asks Link a question and the player has multiple choices for how he answers, so I see that as evidence that in-universe, Link has never been mute.
I think that just like with the Mario Brothers movie having Mario and Luigi talk with new voices, having Link speak in the movie is a concession they would make in service of giving the writers more freedom. Link has spoken before in past media like the Zelda cartoon show, which was part of the Super Mario Bros. Super Show, and since Nintendo is officially acknowledging that as part of Mario's legacy I could see them warming up a little to some of the influence the Zelda cartoon had on the fandom and even the games themselves, within reason.
