Entama has baby and child characters separated by their family, while it's only teens and adults on the V4.
Entama has a cooking feature where you can buy ingredients and combine them to make foods that raise skill points. Entama also has a special secret fourth family that can only be raised by a secret dating code item. Both are absent from the V4. On that note, foods and items are completely different across the two devices.
The way jobs works is pretty different. The Entama just has the Tama going in to and coming back from work. But also, your Tama gets a salary, which increases as you raise the job's respective skill point. On the V4, each job has an associated game, and how often you go to a job and win the game determines the next day payout. Also, the V4 allows you to change professions after a few days, while the Entama locks you into a job once you get it.
Also, the V4 is much more lenient about the requirements to get the three secret characters (Tensaitchi, Makiko, Nonbiritchi), while the Entama is far more strict and requires more work.
Also, while both TamaTown and E-Tamago are no longer active, the Entama's password system was cracked the month it came out, so any item you might want can easily be generated, while the V4's online-exclusive items (especially souvenirs) are currently unobtainable.
On a final note, the V4 can connect and marry with any Connection prior to the V5. The Entama can connect to the Keitai/Akai and Uratama models, but it can ONLY marry another Entama.
