Misty actually is 12 in The Electric Tale of Pikachu manga and sometimes makes a big deal about being older than Ash. Side tangent, but she's also somehow a shotacon in that. Her sisters tease her about being one in chapter 2 and she has a fantasy about Mikey in chapter 6. Like, she gets into it enough that she's pissed at Ash for running by and ruining the mood.
..Anyway, people applied her age there to her anime counterpart, and since anime Misty's already taller and more mature than Ash, it made enough sense for people that 12 stuck.
