Lillie, going solely by what we know about the games, I'd say is the Serena, Dawn, May of Sun and Moon.
Lana is a toss up because we know that she's shy and that she's an action girl. Beyond that we can't be sure.
Lillie for sure will play an active role in the series because her development is going to start with being able to touch and befriend Pokemon properly. Then later on once the Villian arc begins I expect her to become much more involved.
Lana COULD be the regional girl, I just have difficulty seeing it unless she turns out to be a Clemont/Early Serena hybrid. She has two younger sisters so I could see her being more of a "When someone is endanger, her onee-chan instincts kick in to protect them". Like a Oshimizu Nako type character from Hanasaku Iroha. And her development is about getting over her shyness.
I've had a thought about that newest preview with Ash, Kawai and Lana running through the forest. Likely it's Team Rocket causing problems, but what if something happens to Lillie and she winds up getting chased into the forest by a Pokemon or something. Ash comes and saves the day with his Z-Move and protects her which allows Lillie to find a Pokemon, Ashs Pikachu, who she is able to touch for the first time. A lot of anime has a character being able to overcome a fear because they are mentally able to make a connection that if something saves them it's a good thing and that it's not dangerous.
It's likely Pikachu is going to be the first Pokemon she can touch considering how he usually has a key role when Ash meets a female companion, and then the Vulpix at a later date who becomes her first capture and partner.
