It spreads from any Pokemon who has it, whether onset or transferred.

>>15036428 It spreads pretty much immediately, from what I recall. Try having a battle or two. If you keep it out, it becomes inert after 24 hours real time, but putting it in the box pauses the timer. And any Pokemon who catches it can spread it.

Just walking around and fighting wild Pokemon should be enough to start the process. Neither the infector nor infectee have to be in battle.

Any Pokemon with a live virus can spread it to others. If you want to keep the virus live on your mons, put them back in the box before you stop playing your game for the day/night. Spread it to some things that you know you won't unbox, so you always have a live virus ready to spread.
Also, once Pokerus is no longer active, that Pokemon cannot catch it again.
