Those aren't the same. The word for a god or deity, kami, has no implied gender. Kami-sama is the same. Ou is masculine but can be used in a gender-neutral fashion or just to obscure the character's gender.

But the Karate King trainer class in Pokemon is all-male, and in the JP version the Beauty directly references medical science and links it to becoming a Beauty from a Karate King.

It's a Buddhist term referring to the four cardinal gods. In Japanese and other historically Buddhist languages, the term is also used about groups of four people of great importance in a field. These four obviously don't have to be all male just because the word "king" appears in the term.

No, it's just a Japanese term that Japanese people use without really thinking about what it literally means, because that's not relevant in the context it's used in. All languages have shit tons of such words.
