In my opinion, game Serena is our in game avatar (for who play the little girl in games) so she can't give us the right arousal stimulus, because it will be like being aroused by ourselves (ps: I'm aware that this can't be applied to autogynephiliac people).
Anime Serena instead is someone you cannot control, with a specific personality independent from your tastes, someone who can interact with elements around herself, in a different enviroment from the game. So this creates the illusion that she is a "real person" different from her game counterpart, ergo you can be aroused by her.
