Is it possible that the reason that Locke rarely shows positive emotion is that he rarely feels positive emotion?
If so, what would make this happen? Judging by the pictures, there are many attempts by his friends and families to socialize, and they seem to care about him to an extent. He seems to live a rather comfortable, happy life, free of many of the typical stressors an autist would face in our world. Yet, as the expression chart shows, even fucking smiling is a rarity with him. There has to be a reason behind that.
My theory behind it is that as a savant in logical thought, he has considered his role in his life at least a few times. In these, looking at his perception of the world (which, as an autist, would lack the necessary social cues to avert this train of thought), he has come to determine that he is a burden on those around him, and this knowledge plagues him in any and all encounters with those close to him. He isn't happy because he feels like everything he does only harms others, despite the fact that these Pokemon not only do it for free, but they do it for him.
