I remember reading somewhere that a large portion of gen2 designs were canned designs and unused concepts conceived while gen1 was being made. Is there any truth to this?

Yes. Supposedly there's a lot of placeholders in the coding for them, too.

And there was a few gen4 Pokemon that were leftovers that didn't make it into gen3, such as gastrodon.

Gen 1 is the only gen where the index numbers for mons are all over the place instead of being in the order of the pokedex. In doing so, there's a couple of entries in the index list that are "missingno" slots, where if traded to a Gen 2 game they become a Johto mon, which explains stuff like how Ho-oh appeared in the first episode of the anime.
