It's set between the end of the current anime series and the next one which will be set in the region teased in the gen6 games. The game is all about Ash finding the Ho-oh he saw in the first episode of the anime in a Kanto inspired region. It's full of references to the gen1 anime, including Misty and Brock joining you for a while. There's a few references to the gen6 anime as well, with some stuff about Serena and the summer camp thing, but not too much, probably to appeal to genwunners more.
The gameplay is a little different from the main games. The battles, while still turn based, are more dynamic. While types do matter, basic stuff like levels and experience simply isn't there. You only have two stats: Attack and Defense, which are fixed based on the Pokémon you're using. What the moves do and the stats get influenced by how fast you choose what to do and how fast you are at following some instructions on your touch screen during battle. Think of something you'd expect from a rhythm game. There's also some easy puzzle there. Moves, abilities and natures mostly work similarly to the main games. Another thing that affects battles is how Ash and his Pokémon feel. For instance, if you lose a lot of battles everyone gets sad and demotivated, which means they get less defense and they tend to miss attacks more often.
This and Go are probably going to be the biggest releases this year, but I'm pretty sure the next game is going to be announced soon as well.
