In theory it should, but at least in DSi mode or on a real DS/DSi, the system cuts off access to the cartridge slot once it's yanked. This caused problems for some old DS save manager homebrew - you can't just load the save manager, pull the flashcart, put in your game, read the save into RAM, and put back the flashcart to save it to MicroSD. Instead you have to use a slot 2 flashcart on DS, the system SD card on DSi, or FTP on either.
