I think one of the biggest changes is Lusamine who is in her 40s in the games and it is hinted that thanks to Nihilego's poison she stopped aging, perhaps a nod to the butolin toxin from which botox originates. While in the anime she is a normal mother in her late 20s or early 30s similar to Dhelia, even her younger version of her is a clone of Lillie.
