In the anime Mewtwo was made from the DNA found in Mew's eyelash without any implication of what gender the Mew was, and the Adventures version of Mewtwo was also cloned from Mew's eyelash but also had Blaine's genes mixed in making it more male than anything.
Besides, the only reason Femtwo exists is because they couldn't get the rights to first movie Mewtwo for the Genesect movie.
