Technically speaking, saplification starts immediately for non-saplings upon entering /uuu/, its just a matter of time and intensity. As the body's cells mutate, the immune system will naturally get rid of any cells it considers "bad" much the same way it naturally gets rid of cancerous cells when DNA proteins get fucked up during cell reproduction. But because it happens slowly all over your body, after a certain point the genetic edit will be so thorough that it will become the new normal. At that point, theres no going back.
This is also why rapid saplification is so dangerous (outside of the obvious more physical downsides of having leaves grow inside your lungs). A non-sapling's body will be continuously at war with itself trying to fight the saplification of the cells it considers "foreign".
