First of all, the numbers you see there in that image represent a long table, and usually 2 doujin circles share one table together. So a number = 2 circles.
Next, the size of the circles. There are 4 categories that doujin circles are separated to by the Comiket organizer, Small-Medium-Large-Super Large. These are decided based on past sales in the previous comiket, the queue for their doujins, and their initial stock.
Finally, the placement. Similar to size grouping, there are also 4 different placements. Using comiket lingo, they are referred to as Island-Birthday Seat-Wall-Shutter Gate.
>Island (Small/Medium circles)The numbers congregating in the middle of the area, for example E14~E39. Small circles are put into this spot since long queues aren't expected to form from people wanting to buy their doujins, hence it won't lead to congestion in the path between the Islands.
>Birthday Seat (Medium circles)Usually formed impromptu, made out of 4 Islands forming a rectangular shape, for example E13,E15,E38, and E39 if queue is unexpectedly getting longer around those area.
>Wall (Large circles)The numbers on the edge of the convention hall (the wall) like A01 in bottom right spanning through A35 in top left of the image. Those assigned here are expected to have long queues waiting to buy their doujins, which is why they're put on the edge as to not hinder the traffic in the hall by too much.
>Shutter Gate (Super Large circles)If you squint at the image, you can see that on the right of 東1 and top of A28~A30 there's a teal colored line. That is the shutter gate of the convention hall, and the circles put directly in front of it are the heaviest hitters of that particular doujin genre. In this image it's A04~A06, A15~A17, and A28~A30. The reason for this placement is because the queues for these circles are usually absurdly long that it's impossible to contain them inside the hall, so they're are moved outside the hall and only pass through the shutter gate when they finally get to the front line. Holo illustrator like Kanzaki Hiro, Gaou, Hagure, Akasa Ai, and Rin Yuu are placed here. Also remember about 2 circles sharing 1 long table? Those put here usually get to hog the entire table to themselves, sometimes even getting more than 1.
