At the end of the day, it all comes down to making yourself stand out without sounding/looking cringe and desperate. Next step is to make sure you remove any videos/tweets that could very likely make them feel you're not worth the risk. Those could include but not be limited to: political posts, online harassment, lgdhtv topics, strong opinions that shit on anime, vtubers or Japan(-ese) and so on. If your content is focused on that, the chances of you joining Hololive are close to zero.
Another obvious thing, as many anons have already mentioned, is to create content. Music (originals/covers), shorts, skits, streams, art and so on. Being active or/and popular increases your chances because it makes you look like a safer choice. When you have a lot of produced content, they can easily gauge your skill level and decide if you just need a little bit of corpo support to hit it big.
Finally, always keep in mind that no matter how good you are, there's always a possibility they'll give your spot to a relative shitter because that person simply feels more natural for the character they have prepared. That's something multiple JP and EN holos have talked about over the years, so you shouldn't really put yourself down if you don't get in. Be like the JPs who applied multiple times until they finally got in.
My guess is that they'll be looking for 1 musician to be Mori-2 (but not rap), someone for loli/tween chara, 1 MC autist, and someone who has voice acting or drawing experience. There will definitely be 1 big booba chuuba, so if you have that ara ara onee-chan neuron activating voice, I'd say give it a go. 100% they'll look for someone who can speak 2-3 languages, so if you know JP, you're already ahead in the auditions.
