To be a "Reianon" or the more rude version "Reifag" one simply has to like Rei. This is just one of those ways to show that love towards her. There are no initiations for one doesn't need one to be human. It's a matter of will. Without Anno's fantasization and will, Rei would've not been cloathed in the flesh that we can easily distinguish from other anime characters. Rei is Anno's approach to humanity as a whole.
