Basically, the veins can't get 100% of the blood back up out your legs (they're fighting completely against gravity sitting in the average chair/posture), so, they gradually lose oxygen. If you never move your legs, well then blood will clot when it gets super-low on oxygen as its generic response. When you finally move again a big ass internal vein clot can come up and fucking plug your heart or otherwise will get stuck in your lungs (pulmonary embolism which, is also deadly if not treated fast).
In essence humans were designed for walking, and laying down (being plane to gravity negates this risk which is why we don't all die when we fall asleep), and sitting immobilized only for short periods of time. Personally, I have my setup here made so that at least for leisure aka non-gaming on my comp, basically I am usually in a kind of lounged back position in my chair with my feet propped up. I mean normally, people will get up and move enough to generally negate clots just naturally because you wanna go eat or need to piss or something. It's in webcafes where you might lose your spot if you get up that coerces people to just park there for like a day straight and then they die when they get up, this happens sometimes in Asia especially.
