To answer your question. 40km is totally doable for a newbie.
It's going to suck ass if you haven't built yourself up to anything longer than 6-10. You should see what a 15-20km walk does to you in the next couple days. Stretch your legs out afterwards as best you can, to make sure you're not perma sore all the way to your hike night.
You'll want some really nice fitting and padded shoes to help reduce the shock to your feet. Trail runners are my go-to, something with thick foam and good grip. Invest in as good of trekking shoes that you can afford... I've personally clocked in a lot of miles in Altra Olympias, but anything with padding and grip will be a worthwhile investment...
Remember to drink lots of water, and carry some source of electrolytes. The easiest way is to just carry some electrolytes drink packets (make sure it's actually got the essentials, and not just sugar and flavor), or you can rely on food like salty trail mix and fruit. You'll want at least 2 liters of water, make sure you have access to water along the path, or be sure to carry enough to make it.
Assuming that the EKT permits food during the walk, then bring a bag of your favorite trail mix, and a sandwich to get you some much needed calories at the halfway point.
Also bring a dedicated flashlight / headlamp. And a spare battery / charge pack. Getting stuck in the woods at night without a light source really sucks, don't let it happen, trust me.
